Producti~n of a Novel Copper-Binding Ligand by Marine Synechococcus (Cyanobacteria) in Response to Toxic Concentrations of Copper
نویسندگان
چکیده
Marine Synechococcus spp. are extremely sensitive to copper toxicity. Some strains have been shown to produce high-affmity, extracellular ligands of unknown structure which form complexes with free cupric ion. They are also known to produce metaJlothioneins (MT) in response to cadmium and zinc stress. In the present study, marine Synechococcus PCC 73109 (Agmenel/um quadruplicatum BG-1) (Van Baal en) was exposed to three concentrations of CuSO 4 for various times. Size exclusion chromatography, atomic absorption spectrophotometry, and reverse phase HPLC were used to isolate an intracellular copper binding ligand oflow molecular weight ( < 6,500 Da). The ligand was detected after exposure to ~ 8 μM CuSO 4 for 2 hr in BG-11 medium. The intracellular ligand was characterized by electrospray mass spectrometry, amino acid analysis and a universal assay for siderophores. The ligand was not MT, phytochelatin or a siderophore. It is not a peptide but it contains lysine and an unidentified UV 254-absorbing constituent. This compound is a novel copper-binding ligand previously not reported in Synechococcus spp. Key Index Words: copper toxicity, intracellular copper binding ligand, lysinecontained ligand, marine Synechococcus, low molecular weight non-peptide

Coastal strains of marine Synechococcus species exhibit increased tolerance to copper shock and a distinctive transcriptional response relative to those of open-ocean strains.
Copper appears to be influencing the distribution and abundance of phytoplankton in marine environments, and cyanobacteria are thought to be the most sensitive of the phytoplankton groups to copper toxicity. By using growth assays of phylogenetically divergent clades, we found that coastal strains of marine Synechococcus species were more tolerant to copper shock than open-ocean strains. The gl...

Organic chelators that are thought to be of recent biological origin control copper speciation in seawater. Previous studies have often resolved these copper binding ligands into two classes, class 1 ligands (conditional stability constants 1012–1014 M21) and class 2 ligands (conditional stability constants 109–1012 M21). At present the sources and structures of these chelators are unknown. Pre...

The uptake and efflux of 64Cu was studied in the marine cyanobacterium Synechoccous strain WH7803 (DC2). Uptake followed classical Michaelis-Menten type kinetics in metal-buffered seawater. The maximum uptake rate, Vmax, was 0.236 6 0.016 3 10218 mol Cu cell21 h21, with the half-saturation constant, KS, of 10210.8160.11 mol L21. An efflux mechanism was also observed in WH7803, whose growth was ...
